Human FABP3 (Fatty Acid Binding Protein 3, Muscle And Heart) ELISA Kit

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human FABP3. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human FABP3. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human FABP3, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human FABP3 in the samples is then determined by comparing the OD of the samples to the standard curve.

Assay Principle

The Human FABP3 ELISA Kit is based on a Sandwich enzyme immunoassay principle. The microplate wells are pre-coated with an antibody specific to Human FABP3. Standards or samples are added to the wells, followed by a biotin-conjugated antibody specific to Human FABP3. Streptavidin conjugated with Horseradish Peroxidase (HRP) is subsequently added to bind the biotinylated antibody.

After addition of TMB substrate, wells containing Human FABP3 develop a measurable color. The enzymatic reaction is terminated using Stop Reagent, and the optical density is measured at 450 nm ± 10 nm. The concentration of Human FABP3 in unknown samples is calculated by comparing their absorbance values with the generated standard curve.

Assay Workflow

The standard assay workflow consists of sample/standard addition, incubation, washing, addition of biotinylated antibody, incubation, addition of Streptavidin-HRP, further washing, TMB color development and termination of the reaction.

Typically, samples or standards are incubated at 37°C, followed by sequential antibody and enzyme-conjugate incubation steps. TMB substrate is developed in the dark, after which Stop Reagent is added and absorbance is read immediately at 450 nm using a compatible microplate reader.

Performance Characteristics

The kit has demonstrated intra-assay precision of CV <8% and inter-assay precision of CV <10% under the conditions described in the product documentation. Reported recovery ranges were 80–94% for serum, 89–101% for EDTA plasma, and 82–95% for heparin plasma. Linearity testing was also performed using serially diluted serum, EDTA plasma and heparin plasma samples, with the reported results demonstrating acceptable agreement between calculated and expected concentrations across the tested dilution levels. Storage Conditions For unopened kits, short-term storage is specified at 4°C, while long-term storage is specified at -20°C. After opening, certain components including the pre-coated microplate, standard, biotinylated antibody and Streptavidin-HRP may require storage at -20°C, while other reagents are stored at 4°C according to the manufacturer's instructions. Repeated freeze-thaw cycles should be avoided.
