The ELK Biotechnology Human CKMB ELISA Kit is an advanced analytical tool engineered to precisely measure Creatine Kinase MB Isoenzyme concentrations in a variety of biological samples. Please note that this product is for lab research and development only and is not intended for diagnostic use, or use in humans or animals.

Test Principle This kit relies on the Sandwich enzyme immunoassay principle. The provided microtiter plate is pre-coated with an antibody specific to Human CKMB. When standards or samples are added to the wells, they bind to the specific antibodies, followed by the addition of a biotin-conjugated antibody specific to Human CKMB. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is introduced and incubated. The reaction is visualized using a TMB substrate solution, which changes color only in the wells containing Human CKMB, the biotin-conjugated antibody, and the enzyme-conjugated Avidin. The color development is halted with a sulphuric acid Stop Reagent, and the optical density (OD) is measured spectrophotometrically at a wavelength of 450nm ± 10nm.

Kit Storage and Stability

  • If unopened, the whole kit can be stored at 4°C for short-term use (valid for 6 months) or at -20°C for long-term storage (valid for 1 year).
  • Avoid repeated freeze-thaw cycles to maintain reagent integrity.
  • If the kit is opened, store the entire kit at 4°C if it will be used within 1 week.
  • For longer storage of an opened kit (up to 6 months), keep the Pre-Coated Microplate, Standard, Biotinylated Antibody, and Streptavidin-HRP at -20°C, and all other reagents at 4°C.

Sample Collection Guidelines This highly versatile kit supports multiple sample types. Fresh, normal serum or plasma samples are recommended for testing.

  • Serum: Collect in a separator tube, clot for 2 hours at room temperature or overnight at 4°C, and centrifuge at 1000 x g for 20 minutes.
  • Plasma: Use EDTA or heparin as anticoagulants, and centrifuge at 1000 x g and 2-8°C for 15 minutes within 30 minutes of collection.
  • Storage: Assay immediately, or aliquot and store at -20°C or -80°C for later use, strictly avoiding repeated freeze-thaw cycles. Hemolyzed samples should not be used as they will influence the result.
